IT�S HAPPENING: SCIENTISTS CAN NOW REVERSE DNA AGEING IN MICE

Researchers have identified a cellular mechanism that allows them to reverse ageing in mouse

DNA and protect it from future damage.

They�ve shown that by giving a particular compound to older mice, they can activate

the DNA repair process and not only protect against future damage, but repair the existing

effects of ageing. And they�re ready to start testing in humans within six months.

�The cells of the old mice were indistinguishable from the young mice, after just one week of

treatment,� said lead researcher David Sinclair from the University of New South Wales (UNSW)

in Australia and the Harvard Medical School in Boston.

�This is the closest we are to a safe and effective anti-ageing drug that�s perhaps

only three to five years away from being on the market if the trials go well.�

Sinclair and his team made headlines back in 2013 when they found that the cells of

younger mice contained more of a compound called nicotinaminde adenine dinucleotide,

or NAD+, than their older counterparts.

Not only that, but when they gave the older mice more NAD+, they started to look younger,

too.

It was a big deal at the time, but one of the tricky things about medicine is that in

order to show that something could work as a potential treatment, you need to first understand

how it�s acting in the body.

And although the researchers knew NAD+ was having an impressive effect, they couldn�t

say for sure how it was doing it.

Now, Sinclair and his team have released a new study, where they outline in detail the

mechanism through which NAD+ protects DNA from the damage of ageing and radiation in

mice.

So how does it work? When we�re born, all of our cells have the ability to repair DNA

damage, which we experience constantly through random mutations when our cells divide, or

whenever we go out in the sun.

But as we get older, our ability to patch up this damage declines, and our cells being

to age.

What the researchers have now shown in this latest study is that a lot of this damage

comes down to a DNA-repair compound called PARP1.

When there�s a lot of NAD+ in a cell, PARP1 does its job and keeps our DNA healthy. But

when NAD+ drops naturally with age, PARP1 starts to decline, and damage builds up.

To see whether they could take advantage of this cellular mechanism, Sinclair and his

team developed a drug that contains the precursor to NAD+, known as NMN, or nicotinamide mononucleotide.

In mice, boosting older mice with NMN was enough to kick DNA repair into action and

even reverse existing DNA damage.

They now plan to trial a similar drug in humans before the end of the year � and not just

for anti-ageing purposes, but also to protect against DNA damage of any kind.

In fact, the team is collaborating with NASA to see if NMN could help protect its astronauts

against harsh space radiation on their four-year journey to Mars, during which it�s predicted

that 5 percent of the astronauts� cells would die, and their chances of cancer would

approach 100 percent.

The drug could also be useful for groups that are particularly vulnerable to the effects

of radiation, such as frequent fliers and those who undergo frequent CT scans or X-rays.

�The idea is to protect the body from radiation exposure here on earth, either naturally occurring

or doctor-inflicted,� Sinclair told Time.

�If I were going to have an X-ray or a CT scan, I would take NMN beforehand.�

Survivors of childhood cancer could also benefit � right now, 96 percent go on to suffer

a chronic illness by age 45, including cardiovascular disease, Alzheimer�s, or cancers unrelated

to their original cancer.

�All of this adds up to the fact they have accelerated ageing, which is devastating,�

said one of the researchers, Lindsay Wu. �It would be great to do something about that,

and we believe we can with this molecule.�

Before we get too excited, we need to keep in mind that many, many studiesin mice are

not replicated in humans.

So until the results of these early clinical trials in people begin to trickle in, there�s

no promise that NMN will help protect human DNA.

But understanding this mechanism of DNA ageing is a big step towards better understanding

how to keep our cells healthier for longer, and that�s pretty exciting.

Can Bringing Back the Woolly Mammoth Save the Planet?? - Duration: 8:37.

that they thrived in disappeared long

ago transformed by thousands of years of

natural climate change they would have

nowhere to go and you go through all

that work just to put them in zoos seems

go to the zoo to see look at the

researchers think that by cloning a

woolly mammoth and reintroducing it to

its former stomping ground they can help

us stop catastrophic climate change good

ago the world was a very different place

this was the Pleistocene the planet was

much colder by as much as 10 degrees

much of the northern hemisphere lay

buried under miles continent-wide ice

sheets and humanity had just begun its

slow occupation of the world this was

the world of the woolly mammoth the end

of their world in fact with the

recession of the world's glaciers and

the intrusion of human hunting parties

the woolly mammoths would soon be

extinct although a small population hung

on in the northern islands of Siberia

well into the time of the Pharaohs in

Egypt the world they inhabited was

already long gone but there is an

ambitious geoengineering project in

Siberia today with the intention of

resurrecting the woolly mammoth it is

called Pleistocene Park and like the

fictional Jurassic Park its aim is to

bring back an extinct ecosystem much of

Siberia today is made up of dense remote

forests growing in soft mafi ground but

back in the Pleistocene it was grassland

wide open grasslands were the dominant

feature in the Earth's landscape back

then this is where the woolly mammoth

thrived in the intervening millennia as

the climate change these grasslands gave

away to other ecosystems in the lifetime

of our planet ecosystems and habitats

have always been in a state of flux

nothing is permanent far from the

exception extinction and gradual change

are constant and the High Arctic

Siberia is changing again rising global

temperatures are melting the permafrost

present at these latitudes not only is

this phenomenon turning large swaths of

the area into so

drunken forest retrieves begin to list

as if drunk unable to keep themselves

anchored in the newly thawed soil the

warming of the permafrost threatens to

exasperate global warming further by

releasing stores of carbon and methane

trap for eons and the underlying soil

and methane is a much worse greenhouse

gas than co2 it traps about 10 times the

heat energy a mass release of methane

from the once frozen Siberian permafrost

could abruptly accelerate global warming

and this could lead to a hypothetical

event called the clathrate gun a large

reserve of methane can be found frozen

in the Arctic sea bed this is a methane

clathrate basically a form of methane

that is trapped in crystalline ice at

the bottom of the polar seas an

unchecked greenhouse effect could warm

up the oceans enough to melt the ice and

send the methane shooting into the

atmosphere but this ever happened

there'd be no way to stop it like trying

to put a fired bullet back into the

chamber of a gun hence clathrate gun

this has probably happened a couple

times in the Earth's past the largest

extinction event in the history of the

planet the permian-triassic extinction

where ninety-five percent of all life on

Earth perished is thought to have been

caused by a clathrate gun event and

there's evidence of this happening today

in the Yamal Peninsula in Siberia

sinkholes of them found that are most

likely the result of large out gassings

of methane in the melting permafrost but

it's not too late to stop a clathrate

gun from firing all together and that is

exactly what they're trying to do at

Pleistocene Park located in the northern

reaches of Siberia Pleistocene Park has

been slowly trying to reintroduce the

grassland habitat of 12,000 years ago

for 40 years now grassland provides a

much better sink for extracting and

storing greenhouse gases and it has a

much higher albedo meaning it reflects

more sunlight than a typical forest and

thus keeps the planet cooler these

grassland ecosystems may have played a

crucial role in regulating the waves of

glaciation that inundated our planet

over the eons. As the grassland flourished

the planet cooled and when the planet

became too cold the grassland withered

reducing the planets albedo and

releasing its store of carbon back into

the atmosphere allowing the planet to

warm up again but the best way to

maintain a grassland ecosystem is to have a

high density population of large

herbivores along with a variety of other

species the woolly mammoth played a

critical part in cultivating the

planet's vast grasslands during the

pleistocene they had eat tons of

vegetation and that kept rotting plant

material from accumulating and releasing

their carbon into the atmosphere like

their modern-day cousins giant herds of

these massive animals stomp down any

growing trees and kept the forests at

bay for now the researchers at

Pleistocene Park use ATV vehicles and

heavy machinery to crush young trees and

reshape the landscape just as the woolly

mammoths had done bringing them back

could be an efficient way to restore

these vanished grasslands wooly mammoth

bones have been found all over

Pleistocene Park a testament to their

once ubiquity in the area and we've

found woolly mammoth carcasses mummified

in the ice all over Siberia some of them

are so well-preserved we can extract

intact strands of DNA and hypothetically

we could use that DNA to clone them and

bring them back from extinction we've

been cloning mammals for over 20 years

ever since we cloned Dolly the sheep in

1996 and since Dolly we've cloned pigs

horses and bulls so there's no reason to

think we couldn't clone an elephant or a

woolly mammoth but we've never cloned an

animal as old as the woolly mammoth but

there are a few proposed methods we

could extract preserve DNA from a

mammoth carcass and insert it into the

egg cell of its closest living relative

the Asian elephant this would be a

little like growing a chimpanzee inside

of a human mother but we've done this

before the Pyrenean ibex went extinct in

2000 but researchers were able to

extract its DNA and insert it into the

egg cell of a closely related goat three

years after the entire species was

declared extinct the Pyrenean ibex was

born again but the baby only survived

for a few minutes so far we have not

found enough preserved will a mammoth

DNA to make this technique possible so

alternatively if we could find some

preserved sperm from a male mammoth we

could use it to inseminate an Asian

elephant the resulting baby would be a

hybrid but after several generations of

this technique and almost pure woolly

mammoth could be created but frozen

sperm is only viable for about 15 years

we'd have to figure out a way to use

sperm that is thousands of years old

another method would be to modify the

genome of living Asian elephants giving

them woolly mammoth characteristics like

resistance to the cold longer hair and

extra fat this method is already being

tested by Harvard scientists George

church and he successfully inserted

mammoth genes into the Asian elephants

genome although they haven't gone the

extra step and created a viable embryo.

These animals would not be woolly

mammoth they'd be something entirely new

woolly mammoths have been extinct for a

long time how they behaved in the

prehistoric past has been extensively

studied but can't be known for certain

and how they would behave today is not

guaranteed raised by surrogate parents

and scientists these newly resurrected

mammoths would have to adapt to a world

radically transformed from the one they

evolved in while Pleistocene park's

vision is grand and ambitious we might

be too naive to face the immense

complexities of our planets ecology

because drastically altering the earth

landscape to stop global warming is a

little like sticking your finger in the

barrel of a gun to prevent yourself from

being shot it would be easier and much

more likely to succeed if we just take

our finger off the trigger and put the

gun down but if we continue to do so

little to stop the increase of

greenhouse gases in our atmosphere we

may be left with few alternatives one

thing is certain we are changing the

Earth's environment in fundamental and

unpredictable ways the largest global

science experiment in human history is

well underway like the woolly mammoth we

are Pleistocene creatures we came to be

who we are in the same grasslands as

they did and like the woolly mammoths

our past behavior is not necessarily how

we have to behave in the future whether

we are able to correct our mistakes or

adapt to a world radically transformed

from the one that we evolved in will be

the choice we will have to face in the

coming decades--with or without woolly mammoths.

Have you ever fallen for someone you

can't have? Maybe it was someone who was

already in a relationship or married?

Maybe it was someone who you really liked

but they just didn't like you back, or

maybe it was someone who lives so far

away that there was just a very low

chance it was ever going to work out. I

want you to keep watching this video if

that's you because I'm going to talk

about why it is that we sometimes fall

for people that we can't have, secondly

how it can even be self-sabotage

sometimes, and lastly how you can avoid

it going forward. I'm Ben Alexander from

The Legends Academy we helped men with

their confidence dating and

relationships and financial freedom and

today we're talking about why we

sometimes fall for people that we can't

have. Now the reason that this happens is

because you have convinced yourself that

that person that you're falling for is

your only chance or your only option or

your best option. Now let's think about

this for a second just to check if what

I'm saying is true. If you could meet

anyone anywhere (and knowing that on the

whole planet there's so many great

people around), then why would you waste

your time and your energy chasing after

someone that you couldn't have? Now think

about that for a second...why would you

waste all of your time and energy

pursuing someone that doesn't want you

back, or is unavailable, if you could meet

anyone anywhere all the time? Alright so

that's the real problem that's the first

problem. The first problem is although

you might have convinced yourself and

told yourself and said "Oh this is the

only person for me" you know "I've never

connected with anyone this well before"

or it just you know "it's meant to be

it's just she's with the wrong person or

he's with the wrong person" or whatever

it is you're deluding yourself and

telling yourself that they're the only

chance you have out of the whole planet,

all right? Now obviously if you believe

that, you're going to carry on trying to

get someone who doesn't want you back or

that that's already taken, all right? So

that is going to cause you a lot of pain

a lot of agony because you're trying to

force something that's never really

going to work out. Now the second thing I

want to talk about is how it could

sometimes be self-sabotage, now there are

people that on an unconscious level,

meaning that in their mind without them

consciously being aware of it, they're

actually on-purpose looking for

situations where it can never really

work out. There's a very low chance or no

chance that they'll actually ever have

success in that area. And the reason they

do that is if they find someone who's

already taken or live so far away or who

doesn't like them back,

then they can say "oh well it was

never going to happen anyway" so if it

doesn't work out down the track they can

always just say "oh well it wasn't my

fault because you know they were already

married or you know they live so far

away it wasn't my fault it didn't work

out". So they're actually looking for an

excuse or a reason to tell themselves

why you know it doesn't actually work

out so they can sort of let themselves

off the hook in the end. Now the last

thing I want to talk about today is how

you can avoid falling for people that

you can't have. So the first thing to

remember is like what I said at the

start is stop deluding yourself if you

think that that one person is the only

person you ever have a chance with stop

doing that to yourself. The second thing

is if you don't already have the

confidence or the social skills to go

out and meet anyone anywhere, then you

need to do that so you need to find like

a great dating coach or someone like

that that can give you the confidence

and the social skills so you can meet

anyone anywhere. You can meet people

walking down the street, in the

supermarket, at the movie theater, at the

beach, at work, you can meet anyone

anywhere because when you can meet

anyone anywhere, you then completely

massively open up all of your options.

Instead of just having that one option

with that one person, you now can meet,

basically anyone that you can meet is an

option for you now, and if you like them

enough and if you have a good connection

there, then you have the social skills

and the dating skills for example to

move it forward and make it what make

what you want of it. When you can meet

anyone anywhere something really

interesting is going to happen. Because

you're going to be meeting great people

all the time you're going to start to

meet people where you say "oh well that

person I met actually has this thing

that I like a lot better about the other

person" or "this person doesn't have that

thing that kind of used to annoy me

about that person I was hung up on, you

know I've met so many people now that

all have these qualities" and that person

that you are hung up on actually starts

to look like a less attractive option

because you think well "why would I

compromise this and why would I

compromise that when I can just choose

anyone that I want to meet all the time?"

Alright? So you stop limiting yourself to

that one person you open up the whole

world of options you meet all these

other great people and then before you

know it you've usually met at least one

or many more other people that are much

more attractive and a much better fit in

a better option for you than that that

other person that you were kind of stuck

I briefly studied tae kwon do.

And that's when I learned how to use a long staff, a short stick, and nunchucks.

A common pattern is to use rising intonation on all but the last item of a list, as in

one, two, and three.

We use falling intonation on the last item.

Those words are on the beat in English.

The other words - usually little grammar words - they shrink, they get smaller, or link together.

Faking It | February [CC] - Duration: 2:58.

So I kind of think that January and February are the start of the year and if you saw my

January "Faking It" you'll know that January was pretty much a perfect start to the year

in a lot of ways, I felt very excited about what was to come. and inasmuch as it was perfect,

February was the opposite.

It was the worst possible start to the year, my Dad died in February.

and so there's not a lot of content for "Faking It" February, but there is some.

I did some auditions before and some after and I'll show you those now.

First audition of twenty-seventeen!

And I am wearing more makeup than I probably should be, I didn't mean to do that, it's

fine though, it's a theatre audition I don't think that's gonna matter too much.

It's a period piece, and it turns out I am too big for all of the clothes that I would

typically wear to that kind of audition now, so I just picked this dress.

It's kind of 60's style but it's got sleeves, a high neck, it's kind of like, it's a swing

dress and so I think that'll do, that's enough to suggest something a little bit more conservative.

I curled my hair, because I curl my hair all the time now 'cause it looks great, and I

just clipped the top part back to make it look a little bit more 1800''s-y.

Awesome.

Fuck I hate filming in public.

I don't know if you can see me, I'm off to my audition and I didn't have time to film

before I left, the audition is for a commercial.

I kinda like those because you don't really have to do anything, you don't have to prepare

anything, just go in and they give you one line to say or they ask you a question or

whatever it is, so it's a lot less pressure, I think.

I'm not, this is what I'm wearing, I'm not super happy with it.

Basically, a lots happened since last you saw me.

I didn't get either of the two parts from the clips you just saw, which I guess is a

good thing really because I wouldn't have been able to accept them.

Because my Dad died a couple of weeks ago, and so I've just been at my family home doing

all that stuff.

I'm back in Melbourne now, back home, back at it.

I have an audition tomorrow, just found out about it, it's one I self-submitted for, for

a play.

I'd really rather be doing film at this juncture, but, beggars can't be choosers as they say.

So that's it for February.

I don't really know how it feels for y'all to see me not actually getting any parts,

but we'll see.

I have high hopes for March.

The signs of certain diseases or health conditions can sometimes appear on people's faces:

Pale eyelids may indicate anemia, a yellow tinge to the skin could signal jaundice, and

cracked lips may be a warning sign of diabetes.

Now MIT Media Lab spinout Cardiio has developed a mobile app that uses a smartphone camera

to detect facial signs of a heart arrhythmia associated with strokes.

It does so by measuring and analyzing minute changes of light reflected on the skin as

the result of the underlying pulse.

Cardiio is the startup behind the popular fitness app Cardiio that calculates heart

rate based off facial light reflection.

It works because each heartbeat increases the volume of blood in the user's vessels.

Blood absorbs light.

Every time your heart beats, the blood flow to your face and finger increases, causing

more light to be absorbed.

In between beats, less light is absorbed.

By using your smartphone camera to capture tiny changes in reflected light from either

your finger or face, Cardiio App can calculate your heart rate.

Recently, however, the startup has started using that light-reflection data to make a

new app that can detect irregular heartbeat patterns of atrial fibrillation (AF), a common

type of heart arrhythmia that can lead to strokes.

Called Cardiio Rhythm, the clinical app is currently being used for research only and

has been validated by several academic papers.

In the near future, Cardiio hopes to make the app available to consumers for early heart-arrhythmia screening.

Bridging Visa B - The essential facts

We've had articles already about the Bridging Visa A and the Bridging Visa B, but like most

issues about Australian visas it can remain confusing.

Here's a bit of clarity to make life easier for Australian Filipina

couples.

Confusion about Bridging Visa A's (BVA) Might just start with the BVA, even though

this is a fairly easy one.

Why is it easy?

Because whilst technically you do make an application for

a Bridging Visa A, you don't actually complete an application

form , pay a fee and lodge it.

1.

BVA's are free 2.

The application form for all onshore visas that Down Under Visa deals with are also application

forms for a Bridging Visa A. You apply for an onshore partner visa, or onshore tourist

visa?

Or onshore child visa?

Or onshore New Zealand Family Relationship visa?

Your application form is also an application form for a BVA.

It's basically done automatically.

Just please be clear about this bit: Bridging Visas are for onshore visas only!

Offshore visas are not eligible.

So please don't ask if we can get you one!

And be VERY clear about this point: You cannot travel on a BVA!

Leave the country on a BVA, and you are on your own!

You can't return.

Essential points about Bridging Visa B's (BVB) Bridging Visa B's are almost the same as a

BVA.

They allow you to do all the things you can do on a BVA,

with one major difference: You CAN travel on a BVB!

Leave Australia, and abide by the rules of the BVB,

and you can come back again.

Here is what you need to know: * You must apply for a Bridging Visa B

* To get a Bridging Visa B, you must first have a Bridging Visa A!

* The above point?

It means the BVA must be already in place before we can get a BVB.

Don't expect to start on a BVB.

Can't be done.

* A

Bridging Visa B application is like every other visa application, in that you have to

wait for them to grant it.

It's not something you can pick off the shelf in the local Visa Shop.

* And our standard advice of DON'T BOOK A FLIGHT WITHOUT HAVING THE VISA still applies!

Otherwise you may find yourself grounded, and it will not be Down Under Visa's fault.

After the Bridging Visa B (BVB) is granted The Bridging Visa B takes the place of the

Bridging Visa A. So this is now the visa that the applicant holds,

and it remains in place until the main visa application (eg partner visa) is decided.

So no, it doesn't run out.

This is it.

And as I explained before, it's just like a BVA except it allows travel.

Whatever the visa applicant was able to do before, eg. work, study, etc, she may do the

same on the BVB.

No difference there.

What DOES run out is the travel period!

* They will grant a travel period, eg maybe 3 months.

Maybe 2 months.

It depends on the application.

* There will be a date that the visa starts, ie. the date of visa grant

* There will be a "must not arrive after .........." Date.

That means you may travel, but you had better be back in Australia by that date!

* And they will generally be multiple-entry.

So she may travel freely within those dates.

And again, once she gets back to Australia and the date passes, she still holds a Bridging

Visa B.

Maths is all around us.

It's almost impossible to think of something that isn't made without the help of mathematics.

Buildings, transport, technology, medicine-- even clothes and food…

…all rely on numbers, measurements and sums.

We even find maths in the weather outside.

Every type of weather - a cloud floating by, a storm, or the heat of the sun -

can be written down as maths.

Our ancient ancestors used maths to make their forecasts,

along with studying the clouds and the sky.

In the early 1900s in England, Lewis Fry Richardson developed the basic maths we still use

to forecast the weather.

But it was so complicated and took so long that it would have taken around 64,000 mathematicians

all working together to get the forecast done!

Nowadays scientists use super computers, which are so fast, they can do the maths for us

faster than millions of people working together!!

These scientists, called meteorologists, study our atmosphere -

the layer of gases around the earth - and the weather.

All over the world, the weather is measured using instruments and satellites

to help find patterns and make forecasts.

But if you've ever heard anyone complain because the weather forecast was wrong,

you'll know that it isn't as easy as you might think.

This is because weather is a complex system.

The chaos theory or butterfly effect is often used to explain this.

The atmosphere is always changing: even a tiny change, like a butterfly flapping its wings in one place,

can make a HUGE difference to the weather in other parts of the world.

These little changes can happen anywhere at any time.

So the meteorologists worked out even more maths formulas that can include them.

Thanks to these and faster computers, our forecasts are getting much, much better, although

they are still only able to give a good result a few days ahead, not for next month.

The meteorologists don't just give short term weather forecasts;

they study the weather over long periods of time too, to help us learn about climate change.

Climate change is a long term change in the usual weather in an area, such as more rain than before,

or a change in temperature.

While weather can change in a few hours, climate, the normal weather over a large area,

usually takes hundreds or even millions of years to change.

Knowing about the weather is important for growing our food, preparing for severe conditions,

and for helping anyone whose work is affected by the weather, such as sailors and pilots.

It's also really, really handy when planning our spare time and what we're going to wear for the day.
